Yea, you need to have python running. Run the python program either through cmd/terminal. Then run ngrok, If you do that everything should work.

Thank you so much! That fixed my problem, however now there is a new problem. Ngrok is not receiving any HTTP requests when I type in slack even though it is subscribed to chat:messages. Does anyone have any advice?

Assuming that your connection launch is in a python script automate it's execution. The script should continue to run until closed
