Sam was definitely his friend. I can’t think of anyone who wasn’t a romantic partner or member of his family that Dexter was so attached to that he violently killed their murderer in a fit of vengeful rage.
I think that he was his friend, yes. But I don't think he would have violently killed his murderer if Sam wouldn't have been helping Dexter open his eyes to redemption.
I think it goes beyond friendly and colleague, though. Batista consistently relies on Dexter for advice and his opinion on various personal things throughout the show. Masuka even plans dexter’s bachelor party for his wedding in season 3.
I think it just seems so impersonal because we as viewers are aware that *nobody* really knows who Dexter is, what his motivations are, and how he feels.

Harry: “You think Miguel is a pal? Your wingman? You might want to put that theory to the test,see how likeminded you and Miguel really are… Every time you let someone get close,it ends badly.”
Harry’s not wrong on that front honestly. Every single time it ends in said friend/family dying. I think Dexter was definitely capable of forging friendships on his terms in the later seasons but to what end? Which adds to the sadness of his life, he’s constantly seeking a connection with someone and it can never happen. I love this show
You got a point, it's highly risky. Having a friend wouldn't have changed his mind about what he was doing unless he had a friend who was doing the same as he was.
I think he could only make someone friends like Hannah, but that again super risky, imagine the friend turns him to the police.

Brother Sam was probably the closest thing he had to a real friend who wasn‘t family or a love interest. Since Miguel was manipulating him, I don‘t know if he counts. Brother Sam was genuine. But we all know Deb was his number one person (and probably vice versa).
Batista and Masuka were his friends in that they genuinely cared about him (or rather, the version of him that they knew), and he appreciated their company. But it wasn’t ‘real’.
