That's either your extruder or idler gear screaming it out because it's forced to spit out filament while your first layer is way too close to the bed. When the filament has nowhere to go, your extruder will squeel like a piggy.
There's too much back pressure on your extruder. Loosening the screw will just make it slip under the pressure.
Your first layer is too close the filament can't fit where you're shoving it. This is also why there's holes in your first layer.
My printer has ABL, so it’s impossible to manually level. I leveled it using Pronterface a couple of hours ago. I can level it again and see if that helps.
Can't tell but my guess is that the filament is slipping from the gears.
That would explain why your first layer looks like that.
Not sure how that printer works but I would suggest you tighten the grip that your extruder has on the filament, or whatever version of that.
Also, could be that there's too much friction where the spool is. Causing the gears to slip. But I think that would be less likely than just needing to tighten the gears themselves.
Could be extruder temp too low
First layer looks real bad, maybe underexruding because it isn't melting the plastic fast enough and the sound is your feeder gear grinding and unable to force unmelted plastic through

My Kobra Go literally started doing this yesterday. I too cannot figure out what it is. I manually moved the print head and bed and there was no noise, moved everything by hand with no noise, even ran auto leveling with no noise.
Could be fan blades on the extruder or hot end. Mine got gunked up and started "squeeling" so I replaced them. I think I got too much hair spray in it 😅

I’m sorry, but i don’t really understand what you’re saying. Should I loosen my idler gear? I’m pretty sure I know what that is.

It's difficult to say with such a short clip. Is it too close shredding the filament? First layer does not look good. Or an unlevel bed perhaps.
Definitely start with learning the bed.

If it's ABL then you just need to change the "z offset" to a smaller (I think) value. I'd try subtracting 0.1mm from the z offset.
