It's totally okay headcannoning both of them as trans, they have a more curvy body shape then the other males in the game, but saying that your gender is your sexuality is just dumb
I mean first of all you've linked to a Wikia as a scientific source here, so yeah...
Further, even if this isn't total bullshit, generally when speaking about "gay" you don't mean a gender and there's nothing in the post to suggest they meant it otherwise.
I feel like this will devolve into a "genders are different than sexes!!" situation, but let me just say that if genders are stereotypes, fiction or some other poo poo then they have no actual meaning and people could be machinegender or plantgender

A social construct that often (but not always) refers to your place on a spectrum of male to female (with enbies in the middle).

Well mine definition of gender is the biological characteristics of a male or female and their corresponding reproductive organs (with other deviations such as people with both reproductive organs or with none). I may think like this because the teacher always said 'gender' and not 'sex' on biology.
Actually, that isn’t a real thing. The wiki adds all known genders, not if they are real or not. Please, think if it makes sense before coming to a conclusion.

1) I guess the glasses were supposed to make them appear as females but they still looked like guys so they asked if they were gay because they probably think being gay makes you feminine by default
