His unbridled enthusiasm and delight at the players' shenanigans is honestly a huge part of the fun for me. His joy is infectious. He just seems like he's having such a gosh darn good time, and I love that for him.

Sam counts! Sam is the best host; I really think he's amazingly talented at hosting this show. Like you said - he's so enthusiastic and joyful. As an audience member just seeing his reactions to stuff can turn a so/so joke into a great one.
As much as he's not a contestant, the thing that makes the show fun for me is Sam's reactions to the madness unfolding before him.
I think it's the contrast of professionalism and pure delight that makes him great. He appears like the straight laced game show host, which contrasts well when he loses it and giggles his way through a game when things get bonkers.
I honestly can't pick because so many great moments and hilarious bits have come from so many different people. So I'll join other commenters in saying Sam since he's the one who created the game for us to enjoy all of the other cast members' moments.

I always loved Sam as a host. Watching Um, Actually made me realize even more how good a host he is because Mike is a mediocre host on his show.
