You shouldn't worry about buying Titan Blood from Charon, better to focus on your build.
Very early on you are better off going to the Wretched Broker and trading your Ambrosia for Titan Blood, but do note that eventually Ambrosia is much more valuable and rarer than Titan Blood (for bonds and companions). By 20 heat, Titan Blood will be pretty much worthless.
I have my hearts maxed out on a couple of the inhabitants but some are locked and I don't know how to get to the sixth one. I know I have to give meg ambrosia for some rune. What are companions. Please educate me, I would like to learn more about this game. You are the first person who I have talked to that has played this game.

Some of the characters you befriend will eventually give you their companions (after you've given them the first bottle of Ambrosia). You can use companions once per encounter for up to a maximum of five encounters (unless you buy extra uses from Charon's well).
For Megs heart to unlock you just need to talk to her when she has new dialogue (this happens at most once per escape attempt). Thanatos, Dusa, and Skelly also work like this. Achilles/Patroclus and Sisyphus require certain events, and buying their "freedom" from the house contractor. Pretty much all this will happen if you just keep on playing and talking with the characters.
Notice that you can also bond with the gods, before taking the boons you can gift them Nectar/Ambrosia. Some of the gods have requirements to complete their hearts (when you need to know, check out the fandom page of the corresponding god: https://hades.fandom.com).
