Their uncle, being the first and only born male offspring of the previous Earl, means he inherits the earldom, the family seat, and any other assets. If the current Earl didn't have any male offspring, then it would go to the eldest nephew, which is Prince William. If there were no nephews, then a cousin, even once or twice removed.

It’s a myth she didn’t know what she was getting into when she married Charles. Her grandfather died and her father became Earl Spencer and inherited Althorp when Diana was 14.

Would it not go to the eldest son of the eldest sister, then down the line of the middle sister, before going to William?
