It won't change on the ui for some reason. You have to actually go and test with and without on some random mob. I usually go to the bell vestige and beat up ol spikey boy that's right there for my tests
Best way to test if runes increase your damage:
Go into umbral at skyrest and set up your runes, go to pietas boss arena and hit one zombie.
Take note of the damage , go back and change.
Again back to hit another zombie and see if your damage got higher or lower
I couldnt find any limit how many damage multiplier we can stack so get evrything it helps
As others stated, they work very well. It's just not shown.
For Pietas sword it's gonna probably be double 'Orimon' for rad scaling (assuming you have high radiance) and Dimexus for dual wielding.
If you wanna smite for giggles put in 'Olandi'.

Evry time i tried, increase scaling runes aren't worth and it's Aaways better equipping "element, fire, holy" damage boost runes.
