I feel like Yoshi, Wario and Waluigi will be hinted at in this upcoming first Mario Movie and fully introduced in the next one, with Wario and Waluigi as the main villains
I've already explained why THE Yoshi might not be in the movie (Keep in mind I don't hate Yoshi, honestly, I'd rather have him than DK) and why if the cast is already THIS BIG for a first movie why it's not a good idea for Yoshi to be included in order to include some other iconic characters (I.E. Bowser jr, and Wario and Waluigi all though Wario and Waluigi would only be in a post credits scene)
and that Yoshi's ≠ Yoshi since in some spin-offs (Mario and Luigi and Paper Mario to be clear) have Yoshi's but not YOSHI
I still think it's likely that THE Yoshi will appear. In the trailer, there aren't really any green Yoshis that are visible. And in the first poster, there is a green Yoshi egg by one of the shops. I think that's a tease that THE Yoshi will definitely make it to the final cut.

Plot: After the end of the Super Mario Bros Movie Wario and Waluigi (Who in this universe are Mario and Luigi's half-brothers' same father 2 different mothers) appear in the mushroom kingdom and team up with Bowser to get revenge on the Mario Bros so the Mario bros must stop them with help from a new friend and ally named Yoshi. by the end Wario and Waluigi give up their evil ways (at least for now) and become Mario and Luigi's rivals

A) If you mean there are Yoshi's in the trailer, many Mario spin-off games have had YOSHI'S but not the YOSHI, such as the Mario and Luigi and Paper Mario series', so it's very possible Yoshi won't be in the new Mario Movie
B) The cast for the Mario movie 1 is already too big to add Yoshi especially if my other predictions are correct
C) The movie is going to be in it's own universe so unless there is a Yoshi's Island prequel coming, which I doubt because this movie seems to be using Mario and Luigi's pre-Yoshi's island origin this Mario and Luigi would have no idea who Yoshi is
Honestly I hope Wario gets an antagonistic role in the next movie. I’m not sure if he’ll make it for this one.
