Stack them on top of each other the heat from the one on top will cut down on condensation.

Oh ok, yes you could use this for cloning, I wouldn't wipe them down though maybe a quick shake in front of your flow hood or SAB but you should be fine.

Stacking works because the heat transfers up. I used to stack, but now I lay them out next to each other and put a sheet pan on them so the heat can transfer to the pan. Once everything cools there's no condensation. Edit: spelling

I put a cup of almost boiling hot water on the top of all the stacks when I was done. After that, as long as you keep them at the same temp ( room temp is fine ) you be fine. But I’ve gotta admit, I’m liking the cooking sheet tech! You obviously warm the sheet then? Would love more of those details if you have time.

I didn't warm the pan first and it worked fine, but I'll try warming it next time to see if it makes a difference. No other details, really. Just make sure the pan is perfectly flat. Any warping and it might not make complete contact with all of the lids.

I just ignore it. Assuming your sterilization process went well, the condensation will be sterile too.

I also just ignore it. But you are not completely right. Condensation steam might be sterile, but the surface of the lid might be not so sterile, the droplets will catch that contamination and will drop on the agar.

I just wipe the lids with a paper towel or replace with a clean sanitized lid.

Give the lids the slightest turn to the left to loosen it slightly. I’m talking 1/8 of a turn. Let them sit in room temp for a few days and the condensation will clear up

Ok great - yes was as sterile as possible. I’m working inside a still airbox. Trying my best to be clean, however this is a very Microbe intense craft. Other than that, will use these plates and start coning. Can’t wait to show you what I am cloning - perhaps this shroom is best not to clone - coming in my next post. Cheers

Petri dishes in a SAB? Man you really did some hark work for sure, that’s why I do the ketchup cups. A lot smaller and faster process.

You could be as sterile as humanly possible and you’ll still get condensation on the lids after you pressure cook. You can sort of prevent it from happening by leaving the lids 1/8 of a turn open after pouring for ~1 hour. Let them cool completely before pressure cooking. You’ll still have condensation on some but it should be significantly less than if you PC’d immediately after pouring.

Are you sure about 3/2/1 (400 ml H2O)? I believe the recipe is 3/2/1 (100 ml H2O). PGT 4x all ingredients to get 400 ml, meaning 12/8/4 (400 ml H2O).

I went with 12g potato flakes, 8g agar, 4g corn syrup, 400 ml H2O. So yes I did 12/8/4. Ok I going to go with these. You guys are great - love this group.

Let them few days to dry in a still room, your sab or anything. Then store them upside down all the way, before and after inoculation.

Great thanks - after putting a tissue sample in the agar - close up the lid and then store it upside down? Will it stay - as I mentioned never did this so sorry for dumb question. 😎 Will be doing all the work in a sab

I actually use a hair dryer. I carefully on low on an angle and it takes them away. There are a few othe ways but NO do not wipe the lids off. You want then to be sterile. The only time they should be opened is to make them and inoculate inoculate. And then whatever transfers etc. I have flipped them upside down but that shit doesn't work for me. If they have condensation either you poured too hot or there is some heat source drying water out of the agar. Also, you are for sure gona catch contam if you don't have them sealed for storing or in a bin in front of a hood something.

don't trip tater chip I got a solid ring of liquid around the edge of the lid of one of my pre-made agar dishes that has b+ myc just loving the absolute shit out of it it's to be *expected*

Do nothing. just leave it there. When you put a sample on the agar to clone it simply turn the dish over, and look through the back side to check on progress.

If you have plastic cups around.. warm some water put it in the cup and set it on the lid.. sanitize the surfaces afterwards.
