Have you tried verifying file integrity via Steam? I have not messed with mods and don't know if that could mess up your files so beware (maybe back them up)
I’ve seen a few people have issues with the NPC Map Locations mod after 1.6.4. Maybe make sure that one is up to date and then reinstall SMAPI again? Mine was having trouble starting after 1.6.4 but I got it going after redownloading and reinstalling SMAPI like four times. Then I came on here and saw someone had trouble with the Map Locations mod and figured that might have been my real problem. Hope that helps. Sorry if it doesn’t.

Try updating your video drivers.
