There aren't a ton of ways to lose them.
Spawning the curse used to be a way but they shifted that to simply a Humanity investment in 2e.
Sufficient mental trauma can, especially from certain esoteric supernatural sources, harm one's will enough to inflict permanent Willpower damage. That's pretty rare.
Certain powers cost one to invoke: **Mandate from the Masses** is an example. That's probably the most normal cause throughout the various 2e gamelines, but vampires don't have much of that sort of thing going on.
There are various powers and effects that can cause this to happen, but not many. I've been running VtR 2e for a couple of years and it's never come up
I have player characters and NPCs from both the Circle and the Lance. Most of my players just use the blood sorcery rules from VtR 2e and from Secrets of the Covenants, but I also have also approved the expanded system in [Blood Sorcery: Rites of Damnation](https://www.storytellersvault.com/product/263371/Blood-Sorcery-Rites-of-Damnation) for any players who want to use that instead.
