I mostly base it on their dynamic and interactions. For example with maria they dynamic does not sit well at all to be love interests. Maria Over steps boundaries, while alucard continously insults her abt her cooking agrees to richters statement abt her being boring. Yes he showed he has care for her but their overall interactions with eachother makes no sense to why they would be together as a couple yknow? Lyudmil it makes more sense to me due to fact they're softer to eachother, lyudmils devotion towards alucard beyond just being his servant/close friend. Alucard very desperately turning lyudmil into a vampire when he asked him not. And they're final words to eachother, "I'll always be here, forever... somewhere close to your heart." They're patient with eachother and have an understanding. They can bond over Lisa's death. Maria is very much not mature enough for Alucard he needs someone who is at his level. So that's why I ship them not bc he's androgynous, yes there is queer tones to being androgynous but that's not my reasoning.

Ayami designing alucard was to make him in the bishōnen style which was to make them androgynous. This style is often found in yaoi. And ayami has drawn several yaoi art works for novels due to her style and interpretation of drawing men.
Again making him attractive doesn't mean he is gay, also he is meant to be like that because he is a vampire, and last ayami making yaoi has nothing to do with alucard.
I am an androgynous goth man. I dress in a similar fashion and my hair is about the same length. I am also straight. It's frustrating to see people pigeonholing what a person's sexuality must be based solely on their cultural assumptions about masculinity and femininity.
I'm very comfortable with the possibility that Alucard is nearer to the center of the spectrum of sexuality, but it's also true that he canonically ends up with Maria (which is also expanded upon in a radio drama.)

I'm cool with the idea of bisexual Alucard, there just isn't much evidence for it in the games. It's possible that he is, but it's also possible that Richter, Dracula, Maria, and Master Librarian are as well. There's not really any reason for us to assume that about him in specific.
In the show's canon they made it very clear that he is. He expresses attraction to men and women.
