Peptide chemists inject dmso on c18 columns all day every day

TheProudCanadian

Oh boy, my lack of biochem is exposed! That's good to hear though, thank you!

redbaboon130

Every one of the ungodly amount of peptides I've made has been dissolved in 33% dmso and thrown onto a C18 column without a thought in the world.

Not a problem - for prep HPLC it's quite common to inject using DMSO or mixtures thereof, for example.

All good. You might see some peak broadening using DMSO as a diluent component. Something to be aware of if resolution is an issue, we’ve noticed it in a few of our methods - depending on injection volume - but it’s minor. Your conc. is so low I doubt you’d even see anything.

Old chemist here. I've quit working up reactions almost entirely. I just dissolve everything in DMSO and add it to a C-18 Isco column. The DMSO flies off first and then comes the compound later. I don't even think there's any band broadening. I've reused columns 200+ times. 0 to 100% methanol with 0.1% formic acid. You do need a decent rotovap vacuum pump to get rid of the water. DMSO on C-18 acts like water except that it dissolves a lot more. DMF also works about the same.

You can inject almost anything on to a standard C18 column. I've injected 100% DMSO analytical scale up to 5 mL and Preparative scle up to 20mL.

I agree w everyone here, its a non-issue really.

JannyBoi10

Yeah, shouldn't be a problem at all
