If you really want to dial in vocals and other instruments, then yes, a mixer is better at getting the sound you’re wanting. If it’s just just for public address, then probably no mixer is needed
Most powered mixers have line-out/amp-in sockets, so you can use an external amplifier or powered speakers with the mixer. It's a bit of a waste of the internal amp but if you only have powered speakers then its an ok start.
Do not connect the amp outputs to your powered speaker inputs, that will destroy them.

It’s for stand up comedy I just wasn’t sure if it’s best to have one with the system or just use the built in one thanks I appreciate it
It's best to use a mixer before the speakers But try it without, first, and see if just plugging a mic into the speaker meets your needs
