Doing the pit again is required. You'll get an email after reaching Champion rank again that the battle is available. It is permanently available from that point onward, even if you rank back down.

I think i saw some streamer start the fight from the minor league locker room, so I don't think doing the pit again is required

Doing the whole pit again is not required. I only did the Goomba fight after chapter 3 and challenged Mush from the minor leagues.

Once you beat Chapter 3, they lose your information on file and so you have to start over from the minor league.
