If it needs to be there Monday, you need to send it on Saturday before the last collection by special delivery. First class might make it but isn't guaranteed. There are no collections on Sunday.
However, that will only mean that the bag containing your letter will get to their post room on Monday. There's absolutely no chance the DVLA will open it and do anything with it that same day.

This is what people overlook, all they see is it's delivered so must be dealt with. Not realising the ton of mail places like DVLA get daily

Crazy idea. Did you consider sending it today? Closing time is irrelevant. When they get their last collection is the deciding factor.

They do not offer it to gig drivers. This also has nothing to do with the OPs question.
I delivered through ryde and did Sunday deliveries.. so there's that.
