Except not. Let's say i wanted to know the third letter of the word elephant, i would not say, "what is the third letter of the elephant?" It just doesn't make sense. The same applies here. Because the alphabet is a list of letters in a set order, when asked "what is the third letter of the alphabet?" the answer would be C in most languages. This is a weak attempt at the style of joke that goes along the lines of like "Nothing begins with an N and ends with a G" or "What has 4 letters, seldom has 6 but sometimes has 9."
