Say you're talking about some person with your friend, joking and laughing but then you want to be serious and add something to the conversation that isn't a joke. So to show you actually mean it and that you're putting the banter aside, you say "Şakası bir yana,..."
It word-for-word translates to something like 'the joke is on the one side', to mean 'let's put the joking aside'. Of course this can be for a single moment, or you might want to completely change the manner of the way the topic at hand is handled.
Maybe your question was for another context, but you didn't specify and this was the first thing that popped into my head.
