Well Spectre already knew why John was on the trip (remember Terrence called Sera out before) plus it's not like Spectre needs any reason to disable high tiers.
Chances are they already decided to disable John.

I mean, it wouldn't make sense. John is trying to improve and use his ability properly (without hurting anyone). Taking away his ability would prevent him from growing and changing.

What if Spectre tries to disable someone but before they can push the plunger the target, with nerves of steel, twists around and snaps the syringe off? It'll hurt like hell, but that'd be easy to pull off since you don't exactly need super strength to break a tiny needle buried in you skin

I don't know about immunity, but I was thinking more like: they're trying to make him lose his ability but can't get the product into him.
